Genetic relationships of Hemibagrus nemurus in Tasik Chini Biosphere Reserve based on D-loop sequences

Abstract


The population structure of Hemibagrus nemurus in Tasik Chini Biosphere
Reserve was investigated using D-loop DNA sequences. A total of 35 H. nemurus samples were collected from several locations in Tasik Chini and Sg. Pahang for this study. The neighbour-joining and maximum-parsimony analyses based on D-loop sequence data revealed that the H. nemurus samples were clustered into two major clades. The populations from Tasik Chini were grouped together with Sg. Pahang population with 98% bootstrap support. High gene flow (Nm: 2.16-23.3) was recorded among the populations and the pairwise subdivision (Fst) showed no significant value among the populations, indicating no genetic differentiation among the populations. The physical barrier viz. a dam in Sg. Chini does not seem to pose a threat to the genetic diversity of H. nemurus in the Tasik Chini Biosphere.
